In der Fig.4 können wir mit unseren Laborergebnissen erstmals nachweisen, daß im normalen sekretorischen Endometrium gesunder Frauen epitheliales hCG exprimiert wird. Die Genexpression des sekretorischen Endometriums umfaßt sowohl die αCG-Untereinheit als auch die full-length RNA der ßhCG-Untereinheit von Exon 1 bis Exon 3 einschließlich des CTP- Anteils.In Fig. 4 we can demonstrate for the first time with our laboratory results that in the normal secretory endometrium of healthy women epithelial hCG is expressed. Die endometriale RNA wurde extrahiert und durch RT-PCR halbquantitativ zur korrespondierenden GAPDH-Amplifikation bestimmt. Für die Messungen wurden Patientinnen der proliferativen (P, n=22), frühen sekretorischen (ES, n=28), mittleren sekretorischen (MS, n=26) und späten sekretorischen (LS, n=15) Phase des menstruellen Zyklus ausgewählt. In Fig. 6 A ist nach Sequenzierung des ßhCG-Amplifikates eine Nukleotidsequenz für ßhCG ß6, in Fig. 6 B eine Nukleotid-sequenz für ßhCG ß7 und in Fig. 6 C eine Nukleotidsequenz für ßhCG ß7 mit ßhCG ß6 dargestellt. Die Sequenzfolgen bestätigen in hoher Genauigkeit die in Tab. 2 zusammen-gestellten Nukleotidsequenzen für die Expression der endometrialen ßhCG ß7- und ßhCG ß6-Untereinheit. Die ermittelte Nukleotidsequenz basiert auf der Kenntnis des für die Untersuchungen eingesetzte cDNA-Amplifikates mit 548 bp über das Exon 1 und Exon 3. Mit diesen Ergebnissen wird zum ersten Mal eine mRNA-Sequenz für die Expression einer endometrialen ßhCG-Untereinheit vorgestellt.FIG. 6 shows the results of the sequence analysis for confirming the gene expression of βhCG mRNA in the secretory endometrium.
